Project handled: Spinal muscular atrophy (SMA) is an autosomal recessive neuromuscular disease caused by loss or mutation of the survival motor neuron 1 gene SMN1. Objective of the project is to investigate how SMA-linked mutations of SMN1 lead to structural/functional deficiency of SMN macro molecular complex using Saccharomyces cerevisiae as homologous system
Technical Skills:
• Manipulation and tagging of genes in S.cerevisiae with fluoroscent protein genes and affinity tag (STrEP one) using conventional molecular cloning techniques • Purification of native macromolecular SMN protein complexes using Ultracentrifugation and affinity chromatography
• Structural analysis of native macromolecular SMN protein complexes using Cryo EM
• Study of protein - protein interactions using Fluorescent microscope
• Characterization of purified proteins using SDS PAGE and western Blots
Project handled: Investigating the changes in gene expression within the liver of two genetic variants of rainbow trout fish called high stress responders and low stress responders during exposure to a prolonged period of confinement. The obtained transcriptomic profile data can be used for the establishment of biomarker genes against the stress in Rainbow trout.
Technical skills:
• Nucleic acid extractions from different tissues of fish
• cDNA microarrays
• qPCR
• Microarray data analysis using R Bioconductor and GeneSpring software
